A New Glider-Compatible Optical Sensor for Dissolved Organic Matter Measurements: Test Case from the NW Mediterranean Sea ArchiMer
Cyr, Frédéric; Tedetti, Marc; Besson, Florent; Beguery, Laurent; Doglioli, Andrea M.; Petrenko, Anne A.; Goutx, Madeleine.
The MiniFluo-UV is a new glider-compatible optical sensor for measurements of dissolved organic matter (DOM) in natural waters. The working principle, sensor design and challenges faced during the validation phase are reported. The first in situ application of the sensor during three glider deployments in the NW Mediterranean sea (spring, summer, and fall) are also presented. For these campaigns, the two channels of the sensor were adjusted to target Tryptophan-like (excitation/emission wavelengths λEx/λEm: 275/340 nm) and Phenanthrene-like (λEx/λEm: 255/360 nm) fluorescence. These were chosen because they represent fluorophores of interest commonly found in seawater. While Tryptophan (an amino-acid believed to be a by-product of biological activity) is...

Effect of Hydrostatic Pressure on the Fluorescence of Tryptophan in the Presence of Metal Ions Running title: Effect of pressure on the tryptophan BABT
Bao,Chengman; Tang,Xinhui; Ye,Shuming.
ABSTRACT The effect of pressure on the fluorescence of tryptophan in the presence of metal ions was studied by fluorescence spectrometry. It was found that at 60 MPa, the fluorescence intensity of M/Trp mixtures (M represented metal ions) increased compared to that at atmosphere pressure. The relative fluorescence efficiency of M/Trp mixtures increased with pressure. When the M/Trp ratio was above 10:1, the relative fluorescence efficiency in decreasing order was Cu2+/Trp mixtures, Ni2+/Trp mixtures and Mg2+ (K+)/Trp mixtures. When the ratio was below 10:1, the decreasing order was Cu2+/Trp mixtures and Ni2+ (Mg2+, K+)/Trp mixtures. The relative fluorescence efficiency increased with the concentration of Cu2+ and Ni2+. The variation was relate to the...

High grain quality accessions within a maize drought tolerant core collection Scientia Agricola
Ignjatovic-Micic,Dragana; Kostadinovic,Marija; Bozinovic,Sofija; Andjelkovic,Violeta; Vancetovic,Jelena.
Maize (Zea mays L.) landraces are an important source of genes for improving commercial germplasm. Today, drought tolerance and grain quality are major challenges in maize cultivation due to climatic changes and population growth. The Maize Research Institute genebank has a drought tolerant collection, which includes 13 landraces (from the former Yugoslavia) and 12 introduced populations (from different countries). These accessions were analyzed for protein, oil, starch and tryptophan contents, in order to identify drought tolerant accessions with high grain quality. Also, simple sequence repeat (SSR) analysis with specific primers for opaque2 recessive allele (o2) was carried out. All analyzed accessions showed high levels of protein. Oil content ranged...

Modulation of murine blastocyst hatching in vitro by glutamine and tryptophan BJMBR
Castro,W.; Bueno Sánchez,J.C.; Piedrahita-Ochoa,C.; Cadavid J.,A.P..
Enrichment of culture media with amino acids improves embryo development. However, little is known about the specific action of each amino acid during embryogenesis. The present study was undertaken to examine the effect of L-glutamine (Gln) and tryptophan (Trp) on mouse embryo hatching, expansion and viability in vitro. Blastocysts were collected from 6- to 8-week-old female BALB/c mice (N = 30) and cultured in M2 medium containing either 0.125, 0.25 or 0.5 mM Trp, 1 mM Gln, or M2 alone. Gln significantly increased (100%; P < 0.05) blastocyst hatching at 24 h compared to M2 alone or Trp; moreover, Trp inhibited blastocyst hatching when compared to M2 alone (P < 0.05) at 72 h. In contrast, the percentage of embryos reaching the state of expanded...

Nicotine-enhanced oxidation of low-density lipoprotein and its components by myeloperoxidase/H2O2/Cl- system Anais da ABC (AABC)
Oliveira,Olga M.M.F.; Brunetti,Iguatemy L.; Khalil,Najeh M..
In this study, the effect of nicotine on the LDL oxidation by the MPO/H2O2/Cl- system and the effect of HOCl on LDL and some of its components, such as methyl linoleate, vitamin E and the amino acid tryptophan were explored. Nicotine, in micromolar concentrations, enhanced the tryptophan oxidation, either present in LDL or free, in solution. Nicotine also decreased the formation of conjugated dienes and oxygen consumption in a methyl linoleate / HOCl system, and there was evidence to suggest an increase in chlorohydrin formation. Acceleration of the vitamin E oxidation by HOCl was also observed in the presence of nicotine. These data show that the interaction of nicotine and HOCl can promote significant biochemical modifications in LDL particle and some of...

Phenotypic recurrent selection to improve protein quality in non-opaque maize populations Scientia Agricola
Garcia,Antonio Augusto Franco; Souza Jr.,Cláudio Lopes de.
Maize (Zea mays L.) protein is considered to be of low quality due to low levels of the essential lysine and tryptophan amino acids. An alternative to solve this problem is to use the opaque-2 gene, which improves the level of these amino acids, but has negative pleiotropic effects on agronomic characters. A phenotypic recurrent selection scheme was carried out in two non-opaque maize populations to verify the possibility of improving their protein quality without using this gene. Four cycles were completed and a 20% selection intensity for tryptophan content in the kernels was used in two populations, IG-1 and IG-2. The original and the four-cycle populations were evaluated in three locations for agronomic traits. For protein and tryptophan content, a...

Quality protein maize (QPM) contains the opaque-2 gene for twice tryptophan content along with modifying genes for kernel vitreousness. The QPM hybrid needs the inbred lines containing with opaque-2 gene. Therefore, the objectives of this study were to improve the corn inbred lines with high tryptophan by the opaque-2 gene that detected by molecular marker, and to estimate combing ability of these inbred lines for the yield. Eight lines of BC1S5 containing with opaque-2 gene detected by phi057 were used in the experiment. Tryptophan content in the endosperm of 8 selected lines and 28 of their diallel hybrids were analyzed using glyoxylic method. The yield trial experiment was conducted in randomized complete block design with 4 replications. The plot size...
